What PT-141 (bremelanotide) is
The line labelled PT-141 is bremelanotide, a cyclic heptapeptide melanocortin analogue: Ac-Nle-cyclo[Asp-His-D-Phe-Arg-Trp-Lys]-OH, average mass 1025.2. It is Melanotan 2 with a free carboxylic acid at the C-terminus in place of the amide, a difference of a single mass unit, and the lactam ring between the aspartate and lysine side chains is the same.
It is close to MT-2: the average masses are one unit apart, 1025.2 against 1024.2, which tells which compound a batch is; whether a batch also holds some of the other shows on the HPLC purity line, because their isotope peaks overlap. The tryptophan is the light-sensitive residue and the open-chain precursor the synthesis impurity of note. Mid-gradient elution; store dark. The compound is unrelated to triptorelin, which is a GnRH analogue and a separate line on this list.

Identity by mass. Average mass 1025.2 (C50H68N14O10). In electrospray expect the protonated signals near m/z 1026.2 (singly charged), m/z 513.6 (doubly charged); the measured value should sit within about one unit of the calculated one.
- Oxidation: 1 tryptophan, which oxidises to kynurenine and hydroxy forms under light. Expect the largest single related substance to be one of these, eluting just before the main peak.
- Deamidation: no asparagine or glutamine, so the +1 mass deamidation impurity common in longer peptides does not apply.
- N-terminus: capped with an acetyl group, so there is no free N-terminal amine.
- Retention: strongly hydrophilic on the Kyte-Doolittle scale (average -2.20 over 6 residues, standard residues only); the scale alone says little here. Net charge near neutral pH about 0.
